چکیده

Kreuz, Leo E. (The Johns Hopkins University School of Medicine, Baltimore, Md.), and Allan H. Levy. Physical properties of chick interferon. J. Bacteriol. 89:462-469. 1965.-The sedimentation coefficient, diffusion coefficient, and molecular weight of chick-embryo interferon were determined by zone centrifugation and equilibrium sedimentation in cesium chloride density gradients, and by chromatography on Sephadex G-100 columns. Purified interferon is not available in quantities sufficient to permit direct analysis by chemical or physical means; its relative concentrations were determined, therefore, by bioassay. I(125)-human serum albumin was used as an internal reference in all experiments. The sedimentation coefficient of chick-embryo interferon is 2.2 to 2.3S; the diffusion coefficient is 9.5 x 10(-7) cm(2)sec(-1). A molecular weight of 26,000 was calculated from the sedimentation and diffusion coefficients, and a range of 25,000 to 34,000 daltons was obtained from equilibrium-sedimentation analyses.

Viral events necessary for the induction of interferon in chick embryo cells.

Temperature-sensitive mutants of Sindbis virus were employed to investigate the nature of the viral event(s) which induces chick-embryo cells to produce interferon. Chick embryo cells induced by the parental heat-resistant strain of Sindbis virus produced essentially equal amounts of interferon at 29 and 42 C. An RNA(-) and three RNA(+) strains [temperature-sensitive mutants unable (RNA(-)) and...

Effect of ultraviolet irradiation and heating on the interferon-inducing capacity of human adenoviruses.

Heating adenovirus types 8 and 12 for 2 to 3 min. at 560 simultaneously reduced both interferon-inducting capacity and infectivity. In contrast, virus inactivated by u.v.-irradiation effectively stimulated interferon production in chick cells. Chick cells infected with adenovirus type 12 and incubated at 25 °, 35 ° or 4 °o produced about the same amount of interferon but the kinetics of interfe...

A study of events in chick cells infected with human adenovirus type 5 and their relationship to the induction of interferon.

In an attempt to define the molecular events involved in induction of interferon, various parameters of chick cells infected with human adenovirus type 5 were analysed. It was shown by digestion with various proteolytic enzymes and by disruption of the purified virus that induction of interferon requires the interaction of infectious virus with the chick cells. Analysis of adenovirus-infected c...

Effect of interferon on deoxyribonuclease induction in chick fibroblast cultures infected with cowpox virus.

An exonuclease which degrades native deoxyribonucleic acid at pH 9.2 was induced in chick embryo fibroblast cultures and in human amnion cells by infection with cowpox virus. Highly purified chick embryo interferon suppressed the induction of the enzyme in the homologous cell system but not in the human amnion cell cultures. "Mock" interferon prepared from uninfected chicken eggs and purified i...

RNA methylation in vaccinia virus-infected chick embryo fibroblasts treated with homologous interferon.

Interferon-pretreatment of vaccinia-infected chick embryo fibroblasts resulted in a greater than 50% decrease in ribose methylation of the penultimate "cap" nucleotide in virus-specific mRNA. However, in contrast to results obtained with cell-free systems, in intact infected cells there was (a) no detectable reduction in methylation of the 5'-ultimate m7G of viral mRNA; (b) a virus specificity ...
